Converting between TTF/OTF and the web formats WOFF and WOFF2 is lossless: the font data is simply repackaged and compressed. Converting between TTF and OTF changes the outline format (quadratic TrueType vs. cubic PostScript curves); we convert curves precisely and keep all other font tables.
Only convert fonts you're licensed to use. Many commercial licenses restrict web embedding or format changes — check your license first.

WOFF2. It's supported by all modern browsers and is the smallest. Add WOFF only if you need very old browsers.

Converting to WOFF/WOFF2 is lossless. TTF⇄OTF conversion is extremely close but hinting instructions are not carried over, which only matters for small text on some older Windows setups.
Variable fonts can be converted between TTF/WOFF/WOFF2 containers. Converting a variable font's outlines between TTF and OTF isn't supported.
